</font></strong><font style="BACKGROUND-COLOR: #ffffff" size="2"> - 특이적인 항체(antibody, 이하 Ab)로 세포, 조직 내의 target 항원(antigen, 이하 Ag)의 위치와 분포 동정이 목적<br />
- Ab는 형광물질(fluorescent), colorimetric label이 붙어 있어, label 위치로 taget target Ag의 위치를 파악할 수 있는 것임.<br />

Protocol 1<br />
</font><br />
1. 목적 :</strong> cytokine을 monoclonal Ab (단일클론항체)를 이용하여 indirect staning!!<br />
ex) IL-1, IL-4, IL-6, IL-10, IL-12, IFN-r, TNF-α<br />
5) TBS/ Saponin으로 slide washing (3번 * 3분)<br />
6) TBS/ Saponin으로 1/100으로 희석시킨 goat serum을 20분간 처리하여, non-specific한 binding site를 block<br />
7) Ab로 4도씽[서 4도씨에서 overnight incubation<br /> 8) TBS/ Saponin으로 slide washing (4번), biotin이 붙은 secondary Ab(이하 2nd Ab)로 30분간 slide와 incubation<br /> 9) Avidin-biotin-peroxidase를 첨가한 후, distilled water(증류수, 이하 D.W)에 희석시킨 0.5mg/ml DAB solution으로 slide를 incubation시킨다. 그러면 Ag-primary Ab의 정도가 갈색 침전 (precipitate)이 생기게 되어, 이것으로 peroxidase activity를 측정한다.<br /> 10) TBS로 slide washing (4번)<br /> 11) Hematoxylin으로 1분간 counterstaining (착색)<br /> 12) 75% -> 80% -> 100% ethanol 순서로 각각 1분씩 slide를 dehydration<br /> 13) Slide sealing<br /> 14) Optical microscope로 분석<br /><br /><br />
